Our method: exploiting the preferential metabolic habits of tumor cells
Metabolic adaptation is inextricably associated to alterations in cell function. Tumor cells are no exception to this rule. In cancer cells, metabolic reprogramming promotes their growth, survival, proliferation and maintenance. In 1920’s, Otto Heinrich Warburg (who received the Nobel Prize in Medicine in 1931), first demonstrated that cancer cells utilize glycolysis to provide adenosine triphosphate, nucleotide, lipid, and amino acid for their growth, even under aerobic conditions; this phenomenon is called the Warburg effect. Rewiring of energy metabolism was finally elevated to the level of emerging hallmark of cancer by Douglas Hanahan and Robert Weinberg in their extremely influential article Hallmarks of cancer: the next generation, over a century after Warburg’s pivotal finding (Cell, 2000 – Volume 100- 1, p57-70 – Cell, 2011, Volume 144-5, p646-74).
In summary the general characteristics of cancer cells are:

- Cell proliferation involving a higher metabolic activity necessary to produce macromolecules. Rapid cancer cell proliferation, combined with abnormal vascularization structure and function, results in an imbalance in oxygen intake and consumption, leading in hypoxia, forcing cancer cells to rely on glycolysis for energy;
- An increase in defenses against oxidative stress due to a higher production of reactive oxygen forms resulting from this intense metabolic activity;
- Even in normoxia (aka normal oxygen level), cancer cells chose glycolysis as their predominant energy source due to abnormally active oncogene signaling pathways and the tumor microenvironment.

At TheraOnco, we started with the hypothesis that being able to label the Pentose Phosphate Pathway (PPP) would allow us to universally differentiate tumor cells from non-tumor cells, regardless of the type of tumor.
Indeed, the PPP (interconnected with glycolysis), which is « overused » by the metabolism of cancer cells, has the particularity to increasing their defenses against oxidative stress (NADPH Production) as well as ensuring their proliferation via the increased production of macromolecules (production of ribose-5-phosphate – DNA/RNA and erythrose-4-phosphate – aromatic amino acids).
Based on the experience acquired by the CEO of Diamidex (Diamidex – TheraOnco ACA – ACA Project), in the labeling of metabolic pathways in microorganisms via a bio-orthogonal click chemistry approach, we have applied these skills to label the PPP in tumor cells.

Our technology platform is based on localizing bio-orthogonal click chemistry modified sugars in tumour cells via their internal assimilation and metabolization through the PPP. In this process, we make all cancer cells “identical” whatever they were before the assimilation of our modified sugar.
By bio-orthogonal click chemistry – an innovative mechanism that is agnostic to tumour characteristics – we can next covalently link to them all kind of clickable molecules (fluorescent molecules, antitumoral agents, antibodies, nanoliposomes….) allowing us to localise or destroy preferentially tumoral cells.

Conversely, conventional chemotherapies are effective in a large group of patients, but have limited dosing capabilities, lack specificity, and often result in heavy systemic adverse events. The patented technology that TheraOnco have developed, has the potential to bridge this gap.
By using our modified sugars in combination with clickable antitumoral agents we are able to locally concentrate the antitumor molecule where it is needed while keeping its systemic exposure levels below toxic thresholds. The antitumoral agents can be embedded into clickable carriers (e.g. drug loaded nanoparticles), making the technology universal, able to target any type of cancer.
